IGF-1 LR3 (Long R3 IGF-1) is an 83-amino-acid analog of insulin-like growth factor 1, engineered with an N-terminal extension and an arginine substitution at position 3. These modifications were designed to reduce binding to IGF-binding proteins, which normally sequester native IGF-1 and limit its bioavailability — the result is a research tool with prolonged activity in cell-culture systems. In laboratory settings, IGF-1 LR3 is studied in cell-proliferation assays, myoblast differentiation models, and investigations of the PI3K/Akt and MAPK signaling cascades that mediate IGF-1's cellular effects. Because IGF-1 sits downstream of growth hormone in the somatotropic axis, the analog is also used to dissect GH-dependent versus GH-independent growth signaling in model systems. Its extended half-life in vitro makes it practical for multi-day culture experiments where native IGF-1 would degrade.
