GHRP-6 (growth hormone–releasing peptide 6) was the first synthetic hexapeptide shown to stimulate growth hormone release through a receptor distinct from the GHRH receptor — work that ultimately led to the identification of the ghrelin receptor (GHS-R1a). Composed of six amino acids including D-amino-acid substitutions for metabolic stability, GHRP-6 became the prototype for an entire class of ghrelin mimetics that includes ipamorelin and hexarelin. In research settings it is used to probe GHS-R1a pharmacology: receptor activation kinetics, desensitization patterns, and downstream GH-pulse characteristics in animal models.
